背景介紹：This gene encodes a subunit of interleukin 12, a cytokine that acts on T and natural killer cells, and has a broad array of biological activities. Interleukin 12 is a disulfide-linked heterodimer composed of the 40 kD cytokine receptor like subunit encoded by this gene, and a 35 kD subunit encoded by IL12A. This cytokine is expressed by activated macrophages that serve as an essential inducer of Th1 cells development. This cytokine has been found to be important for sustaining a sufficient number of memory/effector Th1 cells to mediate long-term protection to an intracellular pathogen. Overexpression of this gene was observed in the central nervous system of patients with multiple sclerosis (MS), suggesting a role of this cytokine in the pathogenesis of the disease. The promoter polymorphism of this gene has been reported to be associated with the severity of atopic and non-atopic asthma in children.

抗體溶解方法:

方法1：我們的抗體（凍干粉）已經(jīng)包含了0.01M PBS、1% BSA和0.1% 防腐劑（疊氮鈉或*），您只需要加入滅菌的雙蒸水就行了?？贵w溶解后，置于-20℃保存，分裝后使用，避免反復(fù)凍融，可保證至少1年有效；

方法2：也可以只加入一半體積的雙蒸水，再用一半體積的甘油補(bǔ)足，置于-20℃保存，這樣抗體不會(huì)凍，有利于抗體的穩(wěn)定。

后續(xù)試驗(yàn)時(shí)，再使用對應(yīng)的緩沖液稀釋成工作液，即用即配。

我們的抗體（凍干粉），在常溫放置下，至少一個(gè)月有效；若在-20℃下保存（*），至少三年有效。

抗體修復(fù)方式:

修復(fù)液：0.01M 枸櫞酸（pH6.0）或者0.05M EDTA（pH8.0）

抗體修復(fù)方法：

方法1：沸水浴修復(fù)，將盛有修復(fù)液和玻片的燒杯置于沸水浴環(huán)境，保持外部沸騰狀態(tài)15min，自然冷卻至室溫；

方法2：微波修復(fù)，將盛有修復(fù)液和玻片的燒杯置于微波爐中，高火5min，停火3min，中火5min，自然冷卻至室溫。
